Will Plex, Kodi or VLC play the result?
Yes. VLC, Kodi, Plex and Jellyfin all play MKV without extra software. The main place MKV isn't welcome is Apple's own apps — QuickTime Player won't open it, so keep MP4 or MOV for those.
Does converting to MKV lose quality?
Very little. MKV holds the same kind of video MP4 does, so the picture survives the move intact. If you'd rather trade some quality for a smaller file, set a target size or quality level before converting.
